L-norvaline is a non-proteinogenic amino acid, structurally resembling branched-chain amino acids (BCAAs) such as l-isoleucine, l-leucine, and l-valine.[1][2]

L-norvaline belongs to the category of non-proteinogenic amino acids (NPAAs), which are naturally occurring compounds mainly derived from plants and not directly involved in protein synthesis.[1]
L-norvaline functions as an inhibitor of the enzyme arginase, indirectly increasing levels of l-arginine in the body and consequently increasing NO production. It has also been reported to exhibit anti-inflammatory properties and affect signaling pathways involved in inflammation.[1]
L-norvaline serves as a crucial intermediate in the chemical synthesis of Perindopril, an effective antihypertensive drug used to treat high blood pressure.[2]
L-norvaline has been primarily investigated for its potential therapeutic effects, including its role in improving symptoms in various disease models, such as osteoporosis, hypertension, and erectile dysfunction.[1]
L-Norvaline acts as a potent non-competitive arginase inhibitor. It interferes with arginase activity, which is implicated in neurodegenerative diseases like Alzheimer's. L-Norvaline inhibits arginase activity, which helps in restoring nitric oxide synthase activity and L-arginine levels in the brain. It reduces the accumulation of amyloid-beta (Aβ) and tau protein, key pathological hallmarks of Alzheimer's Disease.[3]
